The amino acid sequence of the nitrilase mutant is shown in SEQ ID NO.4, namely, the 167 th tryptophan W of the parent nitrilase is mutated into the glycine G. The nitrile hydrolysis mutant provided by the invention eliminates the hydration activity of parent nitrilase to 2-chloronicotinonitrile, generates no byproduct 2-chloronicotinamide in the catalysis process, greatly improves the nitrile hydrolysis activity, can specifically catalyze the hydrolysis of 2-chloronicotinonitrile to synthesize 2-chloronicotinic acid, and has important potential in the enzymatic industrial synthesis of 2-chloronicotinic acid. |
