The invention provides a graphene/polymer multi-orientation filling modified chemical fiber and a preparation method thereof, and relates to the technical field of fiber modification. In the present invention, in-situ suspension polymerization is used to coat graphene, which greatly improves the dispersion effect of graphene; and comonomer is used to increase the compatibility between graphene and polymer microspheres, so that graphene and PA microspheres are more compatible. A strong interaction is formed between them; the present invention uses graphene/polymer microspheres with low melting point and high toughness to orientately fill the modified chemical fiber, which can increase the strength and toughness of the chemical fiber at the same time, and is expected to improve the electrical and thermal conductivity of chemical fiber products; The graphene/polymer microspheres can form a high aspect ratio microfiber structure inside a single fiber, which can inhibit the formation of cracks in the radial direction (the main fiber fracture direction) on the one hand, and induce polymerization in the chemical fiber matrix on the other hand.
