abstract | The invention discloses a method for constructing a tumor mutation load TMB panel, which comprises the following steps: 1) obtaining transcriptome data of tumor patients, and dividing them into a high TMB group and a low TMB group; 2) screening the high TMB group and the low TMB group Immune-related differentially expressed genes DEGs between groups were enriched and analyzed; 3) Immune cell infiltration analysis was performed by ImmuneCellAI to screen out potential key immune cells; 4) Tumor immune-related genes related to patient prognosis were screened out; 5) Established Modules of different colors and correlation matrix of immune traits that may affect prognosis; 6) Select genes with good correlation with macrophages, DCs, MAIT and infiltration integral in WCGNA, and initially obtain the TMB‑IF gene panel. The invention also provides methods of using the tumor mutational burden TMB panel. The tumor mutation load TMB panel constructed by the present invention has low sequencing cost when used, has low requirements for DNA input, has shorter turnaround time, can perform deeper sequencing, and improves mutation detection sensitivity. |
