titleOfInvention Use of Gedunin in the preparation of medicaments for preventing and treating Huntington's disease
abstract The invention belongs to the technical field of medicine, and relates to new uses of the medicine Gedunin, in particular to the use of the Indian traditional medicine Gedunin in the preparation of medicines for preventing and treating Huntington's disease. The invention uses cell immunofluorescence chemistry, immunoblotting and qRT-PCR to detect the cell line Neuro-2a transfected with mutant huntingtin protein, as well as Huntington patient fibroblasts and Huntington patient inducible pluripotent stem cell orientation from protein level and mRNA level respectively. In differentiated neurons, it was shown that Gedunin degrades mutant huntingtin transfected in the cell line Neuro2a, and endogenous mutant huntingtin in neurons derived from Huntington's patient-derived fibroblasts and induced pluripotent stem cells. Dose- and time-dependent effects of protein aggregates and mutant huntingtin into the nucleus. It can further be used to prepare medicines for preventing and treating Huntington's disease.
