abstract The present invention provides a method for analyzing the genetic structure of wild Salvia miltiorrhiza population by using cpDNA sequence YCF1-RPS15, and the specific steps are as follows: (1) selecting different wild salvia miltiorrhiza populations, and extracting the total genome DNA from young stems and leaves of each wild salvia miltiorrhiza; The primers YCF1–RPS15 were used to amplify the wild Salvia miltiorrhiza population by PCR, and the amplification product of the YCF1–RPS15 sequence was obtained; the primer sequences were: YCF1: 5´CTTGTATGRATCGTTATTGKTTTG 3´; RPS15: 5´CAATTYCAAATGTGAAGTAAGTCTCC 3´; according to the sequence alignment and Bioinformatics analysis was used to obtain the genetic diversity level and population genetic structure of different geographical populations of wild Salvia miltiorrhiza. The method provided by the invention can analyze the genetic variation of the wild Salvia miltiorrhiza population, understand the genetic diversity and genetic structure of the wild salvia miltiorrhiza population in Henan Province, and the results can provide scientific basis for rational utilization and protection of wild salvia miltiorrhiza resources.
