abstract | The invention provides a gene amplification reference substance and application thereof. The method comprises the following steps: and (3) mixing the gene amplification positive DNA with wild type genome DNA according to the following ratio of 1: mixing the S in a volume ratio to obtain an initial amplification positive reference substance; the gene amplification positive DNA refers to a DNA containing only the target gene in which gene amplification has occurred; respectively measuring the amplification multiples of the target gene in the initial amplification positive reference substance and the wild type genome DNA, and respectively marking as X and Y, wherein the amplification multiples refer to the ratio of the copy number of the target gene to the copy number of the reference gene; and (3) mixing the initial amplification positive reference substance with wild type genome DNA at the same DNA mass volume concentration according to the ratio of 1: and mixing the volume ratios of the T to obtain the gene amplification reference product with the amplification fold of Z, wherein Z is any value which satisfies 0< Y < Z < X, and T is S (X-Z)/[ (1+ S) X (Z-Y) ]. The method has the advantages of rapidness, simplicity, low verification cost and strong reproducibility. |
