abstract The invention discloses a special culture medium for nasopharyngeal carcinoma organoids and a scaffold-free culture method. It is composed of double antibody, FBS and Keratinocyte‑SFM. The culture method is to inoculate the isolated nasopharyngeal carcinoma tumor cells into an ultra-low adsorption U-shaped cell culture plate, add a special medium for nasopharyngeal carcinoma organoids, and place them in a 37°C, 5% CO2 incubator after centrifugation. Culture in medium, changing scaffold-free medium every 2-3 days. The special medium for nasopharyngeal carcinoma organoids and the scaffold-free culture method of the present invention do not require the introduction of exogenous scaffolding materials, and can promote cell aggregation into clusters by means of gravity, realize rapid expansion of tumor cells in a short time to construct organoids, and the success rate can reach 100% %, and can maintain the primary tumor specificity; meanwhile, it can maintain long-term tumor cell viability and functional high expression.
