abstract The invention discloses a chromatography medium taking aminobenzene (sulfonamide) pyridine as a functional ligand and application thereof. Taking hydrophilic porous microspheres as a chromatography matrix, and sequentially adding dimethyl sulfoxide and allyl bromide into the chromatography matrix for activation; reacting the activated chromatography matrix with N-bromosuccinimide to perform bromoalcoholization; coupling the chromatography matrix subjected to bromohydrin with aminobenzene (sulfonamide) pyridine ligand; and finally, closing the unreacted bromoalcoholization tail end by adopting an ethanolamine aqueous solution to obtain the mixed-mode chromatography medium taking aminobenzene (sulfonamide) pyridine as a functional group. The novel chromatographic medium can be combined with IgG type broad-spectrum antibody molecules, has the characteristics of large antibody adsorption capacity, high selectivity and salt-independent adsorption, can realize desorption and recovery by changing the pH of the solution to weak acidity, has simple and convenient preparation process and low price, and can be used for chromatographic separation of broad-spectrum IgG type antibodies, including polyclonal antibodies in blood and monoclonal antibody medicaments expressed by cells.
