In this method, the connecting peptide specifically adsorbed on zeolite is first inserted into the plasmid pET28a-bapad vector containing the gene encoding phenolic acid decarboxylase BaPAD through the Nco I/Xho I restriction site, and then pretreated by fusion enzyme The enzyme-immobilized BaPAD catalyst was immobilized on the Na-Y zeolite carrier, wherein the phenolic acid decarboxylase BaPAD was from Bacillus atrophaeus. The biocatalyst prepared by the present invention is used to catalyze FA to produce 4-VG. Compared with whole-cell catalytic FA to produce 4-VG, under the same conditions, the concentration and productivity of 4-VG are increased to 1.97M and 22.8g/L /h; and has good reusability, and can meet the industrial production requirements of 4‑VG. |
