titleOfInvention Construction method and application of an oncolytic adenovirus carrying AIM2 gene
abstract The invention relates to a construction method of an oncolytic adenovirus carrying an AIM2 gene, belonging to the technical field of biomedicine. Firstly, the CAIX promoter vector pZD55-CAIX was constructed by PCR reaction and cloning technology, and then the adenovirus shuttle plasmid pZD55-CAIX-AIM2 was constructed. Finally, the identified plasmid pZD55-CAIX-AIM2 and the adenovirus backbone plasmid pBHGE3 were co-transfected into HEK293 cells, and after the cells appeared viral plaques, the target oncolytic adenovirus Ad-CAIX-AIM2 was obtained. The oncolytic adenovirus can specifically reach tumor cells, efficiently and specifically kill tumor cells without affecting normal cells, and has strong replication ability in tumor tissues.
