titleOfInvention Non-quenching oligonucleotide probe for amplifying variant target gene fragment and application thereof
abstract The invention relates to a non-quenching oligonucleotide probe for amplifying a variant target gene fragment and application thereof, wherein the non-quenching oligonucleotide meets the following conditions: completely matching with a wild target gene fragment and locally mismatching with a variant target gene fragment; adjusting the length of the oligonucleotide probe; modifying at a specific position near the mutation site; fourthly, the probe does not mark a fluorescent group and a quenching group; modifying the 3' tail end of the probe; sixthly, the variation is single or multiple base variation; the probes can be used alone or in combination. The probe balances the inhibition of wild gene amplification and the no influence on the amplification of the variant gene, inhibits the amplification of the wild gene to the maximum limit, avoids influencing the amplification of the variant gene at the same time, and has good enrichment effect on the variant gene. Meanwhile, the method has the advantages of low cost, high flexibility of the enriched detection mode and stable effect.
