abstract | The invention belongs to the technical field of biosensors, and relates to a fluorescence-enhanced sensor for detecting the concentration of Pb 2+ in human serum and a fluorescence analysis method thereof. The method is based on parallel G-quadruplex-induced 4'-aminomethyl-4,5',8-trimethylpsoralen (AMT) fluorescence quenching, and lead ions compete with psoralen for aptamer binding , resulting in the conversion of AMT‑aptamer parallel G‑quadruplex to Pb 2+ ‑aptamer antiparallel G‑quadruplex to release psoralen, constructing a fluorescence-enhanced sensor. Under optimal conditions, the developed aptamer sensor exhibits high selectivity and good accuracy in the range of Pb 2+ concentration from 10 nM to 300 nM, and the limit of detection (LOD) is calculated to be 1.98 nM by the 3σ/K formula. The method is applied to the analysis of human serum for the detection of Pb 2+ by standard addition, which has high sensitivity and selectivity, and has practical application value. |
