The present invention provides a self-contained bifunctional enzyme biocatalyst comprising NADH-dependent specific 3-quinone reductase ( Ml QR), glucose dehydrogenase (GDH) and a flexible linker. The biocatalyst of the invention has the ability of carbonyl reduction and coenzyme in-situ regeneration, the carbonyl reduction activity is 3678U/g, and the coenzyme regeneration activity is 5070U/g, which ensures the recycling of coenzymes and the smooth implementation of the biotransformation process. The yield was 1505.5 g·L -1 d -1 . The present invention is used for the biocatalytic synthesis of (R)-3-quinic alcohol, and can realize carbonyl reduction and coenzyme in-situ regeneration at the same time, without additionally adding expensive coenzyme NADH. The biocatalyst preparation method of the invention is simple in operation, does not need special equipment, has mild process conditions and low cost, and is suitable for industrialization. |
