abstract Methods and compositions for detecting genetic instability using digital amplification assays. The method can be performed in a set of isolated samples, and can generally involve competitive hybridization of competitors and probes/primers to the normal allele and one or more mutant alleles of the microsatellite locus. Competitors can be set up to either compete similarly to the primer/probe or outperform the primer/probe in hybridizing to the normal allele. Primers/probes can be configured to outperform competitors in hybridizing to various mutant alleles of the locus that alter the length of the repeat sequence by varying amounts. Separate aliquots in which primers/probes outcompete competitors can be counted, representing one or more mutant alleles. The method may be capable of diagnosing microsatellite instability and treating a subject based on the diagnosis.
