The invention discloses a construction method of recombinant Yarrowia lipolytica for synthesizing erythritol and a strain thereof. Yarrowia lipolytica was used as the synthetic chassis, and the yeast was genetically edited by means of metabolic engineering improvement to enhance its ability to synthesize erythritol and block the way of synthesizing by-products. The yeast strain is Yarrowia lipolytica ery949-4ΔCGMCC No.19351. Using glucose with an initial fermentation concentration of 50-350 g/L as a carbon source, using a nitrogen source with an initial fermentation concentration of 5-30 g/L and inorganic salts as raw materials, sterilizing at high temperature, and then connecting to the lipolysis method after cooling. Yarrowia strain, continuous fermentation or fed-batch fermentation under aerobic conditions; after fermentation, erythritol is purified from fermentation broth. The strain can efficiently synthesize erythritol by using carbon sources such as glucose as raw materials, without synthesizing by-products (such as mannitol, etc.), and without consuming erythritol. |
