abstract The invention discloses a method for enzymatically synthesizing cinacalcet intermediate (R)-1-(1-naphthyl)ethylamine. The gene sequence and amino acid sequence of the used D-amino acid dehydrogenase are as shown in SEQ ID NO. As shown in 1 and NO.2, it can be prepared by constructing Escherichia coli genetic engineering bacteria and fermenting culture in large quantities; in the potassium phosphate buffer of pH6.5~8.5, add 50~300mmol/L 1-naphthoethyl ketone concentration respectively, 100~ 600mmol/L amino donor, 0.1-0.5g/L coenzyme, 5%-10% (V/V) co-solvent, 1-3g/L coenzyme cycle enzyme, 0.5-2g/LD-amino acid dehydrogenase, at 30 The reaction is carried out at ~40° C. for 24 to 48 hours to generate (R)‑1‑(1‑naphthyl)ethylamine, the final reaction conversion rate is greater than 95%, the product yield is greater than 88%, and the product chiral purity reaches 99% ee. The invention has mild reaction conditions, small amount of organic solvent, greener reaction system and high conversion rate, greatly improves the preparation efficiency of (R)-1-(1-naphthyl)ethylamine, and has good industrial application prospects.
