abstract | The invention provides a method for distinguishing lacidipine individualized medication by mass spectrometry through a detection product, which utilizes extension primers with different Single Nucleotide Polymorphism (SNP) sites with different molecular weights to simultaneously detect a plurality of sites related to lacidipine metabolism of antihypertensive drugs by matrix assisted laser desorption ionization time of flight mass spectrometry (MALDI-TOF MS), and comprises the following steps: respectively designing a multiplex amplification primer and an extension primer according to 6 target SNP sites to be detected; preparing a multiple amplification primer reaction system and an extension primer reaction system; in a reaction system, a plurality of sets of primers are used for simultaneously and respectively carrying out amplification and single base extension reaction on 6 target SNP sites; and (3) performing flight time mass spectrometry on the product obtained after the single base extension reaction, identifying the genotype of SNP (single nucleotide polymorphism) related to different drug metabolism according to the products of the extension primers with different molecular weights represented by mass spectrum peaks, and guiding the administration of the antihypertensive drug lacidipine. The invention can simultaneously detect 6 SNP sites related to metabolism of lacidipine, which is a antihypertensive drug, and has the advantages of low cost, no need of synthesizing probes, short time consumption, simple and convenient result analysis and extremely wide application field. |
