abstract The invention relates to a ratio fluorescent probe for detecting PTK7 and a preparation method thereof. The probe adopts Fe 3 O 4 nanoparticles as a quencher and a magnetic separation agent to cross-link the complement of PTK7 aptamer (APT) on the surface. The strand (cDNA) specifically binds to APT labeled with yellow fluorescent emitting carbon dots (y‑CDs) and quenches its fluorescence. PTK7 competes with cDNA to bind APT to make it detached from Fe 3 O 4 nanoparticles to restore fluorescence, and is filtered by fluorescence internal filtration. The effect quenches blue fluorescence-emitting carbon dots (b‑CDs), resulting in ratiometric fluorescence detection of PTK7. Exonuclease I increases the fluorescence ratio of y-CDs and b-CDs, and the ratio of the two fluorescence intensities I 580 /I 460 is linear with the concentration log10 of the analyte. According to the ratio of the two fluorescence intensities, quantitative determination of PTK7 Biomolecules with high sensitivity, simple detection, high selectivity and high affinity of immune response.
