titleOfInvention | Prokaryotic promoter report system based on lacZ gene and pUC replicon, and construction method and application thereof |
abstract | The invention discloses a prokaryotic promoter reporter system based on lacZ gene and pUC replicon, which comprises pUC replicon derived from pFLX107, Cat gene, rrnbT1T2, lambda T0 transcription terminator, lacZ gene derived from plasmid pRCL and with silent mutation of internal ClaI, SacI and NdeI sites. The invention also discloses a construction method and application of the prokaryotic promoter report system. The size of the plasmid pFGH06 constructed by the invention is 4949bp, the background activity under the culture condition of 28 ℃ is only 12.2 +/-0.6, and is extremely obviously lower than the background activity of 21.3 +/-1.7 of a low-copy reference plasmid pRCL, the plasmid pFGH06 is applied to cloning and activity determination of an inducible promoter araBAD and a constitutive promoter rpsM, and when the plasmid pFGH06 is simulatively applied to promoter screening, 100% identification of a target promoter can be realized through blue-white spot screening. |
