titleOfInvention Oocyte immunofluorescence staining method based on hydrogel wrapping technology
abstract The invention belongs to the field of cell staining, and particularly relates to an oocyte immunofluorescence staining method based on a hydrogel wrapping technology. The method comprises the following steps: transferring the oocyte to an enzyme-labeled hole, dropwise adding hydrogel to cover the oocyte, and performing photocuring; subsequently, 4% paraformaldehyde fixation, Triton-X100 permeabilization, 1% BSA or goat serum blocking were performed, followed by primary antibody incubation, secondary antibody incubation, DAPI staining. Aiming at the oocyte which is large in size, limited in quantity, non-adherent and non-proliferative, the hydrogel packaging technology is adopted for carrying out cell immunofluorescence dyeing, the hydrogel has good biocompatibility and permeability, the whole immunofluorescence dyeing process is carried out in an enzyme labeling hole sealed by the hydrogel, the problems of oocyte loss, complex operation, low efficiency and the like caused by multiple transfer of a mouth suction tube in the traditional oocyte immunofluorescence dyeing process are solved, the dyeing effect is good, and the method is very suitable for popularization.
