abstract | The invention discloses a new application of a pig GADD45a gene and construction and application of a high-expression cell line. Designing and synthesizing a GADD45a full-length amplification primer containing a restriction enzyme cutting site according to a pig GADD45a gene full-length sequence and a vector pLEX-MCS; amplifying a GADD45a gene full-length sequence from a piglet adipose tissue, recovering and carrying out enzyme digestion, connecting the sequence with the enzyme digested pLEX-MCS, constructing a pLEX-MCS-GADD45a lentiviral plasmid, carrying out monoclonal sequencing identification and verification, and then co-transfecting the pLEX-MCS-GADD45a lentiviral plasmid, pSPAX.2 and pMD2.G into 293T cells by adopting a liposome transfection method to obtain a recombinant lentivirus; transfecting a pig IPEC-J2 cell by using the recombinant lentivirus obtained in the step 2) to obtain an IPEC-J2 cell line of a high-expression pig GADD45a gene. The new application of the pig GADD45a gene is used for regulating and controlling nutrition absorption and metabolism. The invention develops a new function of the pig GADD45a gene and a new method for researching the same in intestinal epithelial cells. |
