In this method, a three-dimensional dynamic culture environment was constructed by using a rotary wall bioreactor (RCCS), and FACTâ…¢ microcarriers were selected as scaffold materials to establish a three-dimensional dynamic culture system that could support the rapid proliferation of mouse testis SSCs in vitro and maintain their undifferentiated characteristics. Under this three-dimensional dynamic culture system, SSCs showed excellent proliferation effect after being cultured in RCCS for one week, especially the SSCs obtained in vitro for 17-22 days proliferated rapidly, and the proliferated SSCs still retained their undifferentiated biological properties Features and Functions. The method of the invention has obvious advantages such as simple operation, low culture cost, good repeatability, high culture success rate, no need for passage, and avoidance of feeder layer cell contamination, and provides a reliable technical solution for in vitro culture of SSC.
