titleOfInvention | Chinese rose polysaccharide, extraction method and application thereof in preparation of anticoagulant drugs |
abstract | The invention provides a method for extracting Chinese rose polysaccharide, which comprises the following steps: sequentially extracting dried China rose with petroleum ether and 90-95% ethanol solution at room temperature, filtering after extraction, extracting residues with distilled water, concentrating the filtrate under reduced pressure, then precipitating with ethanol, collecting precipitate, removing protein by Sevage method after redissolving, concentrating the supernatant under reduced pressure, precipitating with ethanol again, sequentially washing the precipitate with diethyl ether, acetone and ethanol, and freeze-drying the precipitate to obtain China rose crude polysaccharide; purifying the crude polysaccharide of the Chinese rose by DEAE-52 anion exchange column chromatography, DEAE-Sepharose FF chromatographic column purification and Sephadex G-100 chromatographic column further to obtain the Chinese rose essential polysaccharide RCJ 2-Ib. The invention inspects the influence of RCJ2-1b on various biochemical indexes of an acute blood stasis model rat to investigate the anticoagulant activity of the rat. The test results prove that: the Chinese rose polysaccharide RCJ2-Ib has a remarkable in-vivo anticoagulant effect and can be used as a potential anticoagulant drug. |
