abstract | The invention relates to a preparation method of total saponins of emblic leafflower fruit, in particular to a method for extracting the total saponins of emblic leafflower fruit by adopting an enzyme method-ultrasonic wall-breaking method. The processing steps comprise: preparing emblic leafflower fruit, crushing and grinding the emblic leafflower fruit into powder, adding citric acid-disodium hydrogen phosphate buffer solution with the pH value of 4.5 according to the material-liquid ratio of 1:8, uniformly mixing, adding 5-20% of cellulase and hemicellulase mixed enzyme solution according to the mass, carrying out enzymolysis for 1-2 h at 50 ℃, centrifuging for 10min at 4000r/min, collecting supernatant, taking a proper amount of enzyme-method wall-broken precipitate, respectively adding warm water, 40-60% of absolute ethyl alcohol and 0.08mol/L of citric acid-disodium hydrogen phosphate buffer solution with the pH value of 3.8 according to the material-liquid ratio of 1:8, extracting for 60-120 min, carrying out ultrasonic assisted extraction under the same conditions, after extraction is finished, centrifuging for 10min at 4000r/min, taking supernatant, combining the supernatants, and drying at 80 ℃ to obtain the emblic leafflower fruit total saponin. The invention has the advantages of high efficiency, high content of effective components in the solution and strong oxidation resistance. |
