abstract | The invention provides a method and a kit for guiding the administration of levamlodipine for a hypertension-reducing medicament levamlodipine administration guide gene detection kit. The method comprises the following steps: respectively designing a multiplex amplification primer and an extension primer according to 4 target SNP sites to be detected; preparing a multiple amplification primer reaction system and an extension primer reaction system; in a reaction system, a plurality of sets of primers are used for simultaneously and respectively carrying out amplification and single base extension reaction on 4 target SNP sites; and (3) performing flight time mass spectrometry on the product obtained after the single base extension reaction, identifying the genotype of SNP (single nucleotide polymorphism) related to the metabolism of different drugs according to the products of the extension primers with different molecular weights represented by mass spectrum peaks, and guiding the administration of the antihypertensive drug, namely the levamlodipine. Also provided is a detection kit using the method. The kit can simultaneously detect 4 SNP sites related to the metabolism of the antihypertensive drug L-amlodipine, and has the advantages of low cost, no need of synthesizing probes, short time consumption, simple and convenient result analysis and extremely wide application field. |
