titleOfInvention A kind of determination method of trace chlorophyll in high organic matter soil
abstract The invention discloses a method for measuring trace chlorophyll in high organic matter soil, comprising the following steps: S1, sampling: selecting soil samples and green plant samples to be tested, and freeze-drying; S2, extracting chlorophyll; S3, using ultraviolet-ray Visible spectrophotometry was used to determine the optical density of the green plant chlorophyll extract at wavelengths of 663 nm and 645 nm, and the Lichtenthaler method was used to calculate the content of chlorophyll a in the green plant chlorophyll extract; S4, three-dimensional fluorescence spectrum determination; S5, draw a standard curve , establish a regression equation; S6. Determination of the content of chlorophyll in the soil to be tested. The invention utilizes the low-background three-dimensional fluorescence spectrum to solve the problem that trace chlorophyll in the visible ultraviolet spectrum is greatly interfered by colored backgrounds such as peat, high organic matter soil, etc. The method of the present invention can be widely used to study the process of green plant litter and humification.
