abstract The invention discloses a preparation method of letrozole, which comprises the following steps: adding p-cyanobenzaldehyde and triazole into a solvent, and reacting to obtain 4- (1- (1H-1,2, 4-triazole) hydroxymethyl) benzonitrile; adding thionyl chloride to the above product; adding the product and tert-butyl alcohol into a solvent, adding a catalyst, and reacting to obtain 4- (1- (1H-1,2, 4-triazole) chloromethyl) benzoyl tert-butylamine; adding the product into a solvent and a Grignard reagent, and reacting to obtain 4,4' - ((1H-1, 2, 4-triazole-1-yl) methyl) bis (N- (tert-butyl) benzamide); adding phosphorus oxychloride, and reacting to obtain letrozole. The reaction routes of the invention are mild, do not relate to reactions under harsh conditions, do not relate to high-risk processes, and are suitable for industrialization.
