abstract The invention provides a primer composition for distinguishing individual medicine type of antihypertensive medicine captopril, wherein the preparation method comprises the following steps: respectively designing a multiple amplification primer and an extension primer according to a plurality of target SNP sites to be detected; preparing a multiple amplification primer reaction system and an extension primer reaction system; in a reaction system, a plurality of sets of primers are used for simultaneously and respectively carrying out amplification and single base extension reaction on a plurality of target SNP sites; and (3) carrying out flight time mass spectrometry on the product obtained after the single base extension reaction, identifying the genotype of SNP (single nucleotide polymorphism) related to different drug metabolism according to the products of the extension primers with different molecular weights represented by mass spectrum peaks, and guiding the individual administration of the antihypertensive drug captopril. The invention can simultaneously detect 5 SNP sites related to captopril drug metabolism, and has the advantages of low cost, no need of synthesizing probes, short time consumption, simple and convenient result analysis and extremely wide application field.
