abstract | The invention discloses a vector for inhibiting geminivirus infection based on a CRISPR/Cas9 SunTag system, and construction and application thereof, wherein the method utilizes the CRISPR/dCas9 Sun Tag system carrying methyltransferase DRM2 to construct sgRNA aiming at specific regions of TYLCCNV and TYLCCNB, so that the specific regions of TYLCCNV and TYLCCNB are identified by DRM2 and methylated. In order to enhance the recognition and methylation effects, methylation sensitive regions of three sgRNAs (g16/g17/g9A) as TYLCCNV targets and three sgRNAs (g1/g14/g9B) as TYLCCNB targets are respectively designed, and the three sgRNAs are sequentially inserted into a pEG302 vector, so that a vector capable of inhibiting geminivirus infection, which can target a site and perform methylation modification on pEG302-A of TYLCCNV, and target a site and perform methylation modification on pEG 302-beta of TYLCCNB, is obtained. |
