abstract | The invention discloses an extraction method of total flavonoids in neem leaves, which comprises the following steps: reflux-extracting folium Azadirachta indica powder with 70-80% ethanol for 2-4 times (each for 2-3 hr), filtering the extractive solution, evaporating to dry, dissolving the residue with hot water, extracting with petroleum ether solution, discarding petroleum ether solution, concentrating the aqueous solution in an evaporation dish to obtain extract; dissolving the extract with appropriate amount of water, enriching with macroporous resin, washing with 10-20% ethanol for 7-9 column volumes to remove impurities, eluting with 75-85% ethanol for 10-12 column volumes, collecting 75-85% ethanol eluate, separating by silica gel column chromatography, eluting with chloroform-methanol at volume ratio of 75:1, 45:1, 22:1, and 5:1, collecting 22:1 eluate, and concentrating. The method can effectively extract total flavone components from folium Azadirachta indica, has effects of reducing blood sugar, resisting bacteria and virus, and can be used for large-scale preparation. |
