abstract The invention discloses a method for optimizing a carbon source of a staphylococcus carnosus high-density culture medium and application thereof. The carbon source that staphylococcus carnosus can utilize was comprehensively screened from 95 different carbon sources, colloids and tetrazoles contained in the identification plate by a full-automatic microbial identification system (Biolog) method. Based on the principle of color reaction, the instrument reads the absorbance at 590 nm. In the Biolog method, a single colony of bacteria to be detected is purified by using a BUG solid plate, then the single colony is picked by using a cotton swab and uniformly mixed in inoculation liquid containing gram-positive bacteria, the bacteria are diluted, then the diluted bacteria liquid is sucked by using a discharging gun and inoculated on a GP2 positive 96 pore plate, continuous culture is carried out for 15 hours, reading is carried out every 1 hour, then the discoloration condition of a culture hole is observed, a carbon source capable of being utilized by the bacteria is judged, then statistical analysis is carried out on specific numerical values, a high-quality carbon source capable of being utilized by staphylococcus carnosus is selected by using visual numerical values, and then high-density culture is carried out by using the carbon source.
