abstract | The invention discloses a method for extracting filamentous fungus total RNA from yeast materials, which comprises the following steps: 1) pretreatment: screening a solid-state fermentation yeast sample with complete and full raw material particles, and freezing; 2) primary grinding: adding the spherical grinding aid into the solid fermentation starter material obtained in the step 1), adding liquid nitrogen, grinding to strip and drop thalli on the surface of the starter material, and simultaneously keeping the integrity of raw material particles in the starter material; 3) after primary grinding, removing raw material particles to obtain a thallus mixture; 4) fully grinding the thallus mixture obtained in the step 3) to obtain powder material containing thallus RNA; 5) adding a total RNA extractant into the powder material obtained in the step 4) to extract total RNA, thus obtaining the RNA-RNA composite material. The method is suitable for extracting the total RNA of the filamentous fungi from complex matrixes such as yeast and the like, and the total RNA of the filamentous fungi extracted by the method has high purity and good integrity. |
