abstract The invention provides a preparation method of fingolimod hydrochloride. The method uses 2,2-disubstituent-5-hydroxymethyl-5-nitro-1,3-dioxane to react with halogenated reagent and triphenylphosphine in turn to prepare corresponding Witting reagent, and then react with para-n Octylbenzaldehyde is subjected to Witting reaction, catalytic hydrogenation to reduce nitro group and carbon-carbon double bond, hydrolysis reaction to remove hydroxyl protecting group aldehyde or ketone to prepare fingolimod, and then salify with hydrogen chloride to obtain fingolimod hydrochloride. The method of the invention has high reaction selectivity and yield, cheap and easy-to-obtain raw materials, high safety and operability, mild reaction conditions, less waste water generation, and the obtained fingolimod has high purity and is high-purity fingolimod hydrochloric acid. The preparation of the salt provides a guarantee for the easy and industrial production of fingolimod hydrochloride.
