abstract | The invention discloses a method for quantifying non-enzymatic modification sites of hemoglobin by using LC-PRM/MS and its application. The method includes: 1) incubating a small molecule standard solution with hemoglobin in vitro to obtain a modified hemoglobin sample in vitro; 2) extracting hemoglobin in human blood; 3) using LC-MS/MS to separate trypsin, chymotrypsin and hemoglobin, respectively. The in vitro and in vivo hemoglobin peptides obtained after the enzymatic hydrolysis of Staphylococcus aureus V8 protease were detected; 4) the information distribution of hemoglobin modification sites was obtained from the detection results and the modification site spectral library was established; 5) the hemoglobin after enzymatic hydrolysis was Peptide fragments were mixed for monitoring experiments; 6) According to the modified site spectrum library, LC-PRM/MS was used to screen peptide markers for early diagnosis of diabetes.
