abstract | The invention discloses an expansion method of stem cell-like memory T cells, which comprises the following steps: step one, separating lymphocytes from peripheral blood or umbilical cord blood by using lymphocyte separation liquid; secondly, resuspending the lymphocytes with the first solution, performing amplification culture on the TSCM cells with the second solution after 24 hours, and obtaining the TSCM which reaches the application standard after 22 days of amplification culture; the formulation of the first solution comprises: RPMI1640 medium, HEPES: 25mmol/L, penicillin: 100U/ml, glutamine: 2mmol/L, fetal bovine serum: 9-10% (w/v); the formulation of the second solution comprises 1-4U/ml of cytokine; the invention solves the problem that memory T Cells (TCM) are easy to be differentiated into TEM in the in vitro culture process; the induction activation rate of the stem cell-like memory T cells (TSCM) is high. |
