abstract | The invention relates to the field of plant antiviral gene engineering, and discloses a potato Y virus gene fragment for efficiently generating siRNA, an attenuated vaccine, a preparation method and application thereof. The potato Y virus gene segment for efficiently generating siRNA comprises at least one of a PVY1 segment, a PVY2 segment and a PVY3 segment, and the nucleotide sequences of the PVY1 segment, the PVY2 segment and the PVY3 segment are respectively shown in Seq ID No.13, Seq ID No.14 and Seq ID No.15, and the gene segment can efficiently generate siRNA after being inoculated with a parasitic plant. The attenuated vaccine is based on TVBMV attenuated mutant, the TVBMV attenuated mutant is embedded with effective gene segment which can induce the potato Y virus to generate cross protection, and the effective gene segment comprises potato Y virus gene segment which can generate siRNA. The potato virus Y-resistant attenuated vaccine has stable effect, can play an effective cross protection role, remarkably reduces the damage of plants infected by potato virus Y virulent strains, delays the onset of the plants and greatly reduces the loss. |
