abstract | The invention provides a method for preparing retinal ganglion cells by in vitro reprogramming, which comprises the following steps: obtaining somatic cells integrated with an Ascl1 gene, an Islet1 gene and a Brn3b gene on a chromosome; performing induction culture to obtain retinal ganglion cells; the induction culture comprises the following steps: inoculating the somatic cells into an MEF culture medium added with doxycycline to be cultured for 1-3 days; then, the culture is changed into a mixed culture medium of DMEM/F12 added with doxycycline and a nerve cell culture medium for 11-13 days; and, FGF2 was added to the medium on days 1 to 5 from the start of the induction culture. In the reprogramming process, the cell growth factor FGF2 is selected and added to the induction culture medium, and particularly, the cell growth factor FGF2 is added at a proper time, so that the reprogramming efficiency can reach more than 30%. |
