The invention relates to the field of stem cells and regenerative medicine, in particular to a method for researching the dryness of mesenchymal stem cells without a scaffold three-dimensional gel. The mesenchymal stem cells were subjected to gel induction and culture for 7 days to obtain a scaffold-free three-dimensional gel of mesenchymal stem cells, which were respectively inoculated into adipogenic, osteogenic or chondrogenic induction media for 14 days of induction and culture, and the medium was changed every 72-96 h. The environment was a 37°C, 5% CO2 incubator. After differentiation induction, oil red O was used for adipogenic staining, alizarin red for osteogenesis, and Alician blue for chondrogenesis. The invention provides a method for verifying that the mesenchymal stem cell gel obtained by drug induction still maintains the multidirectional differentiation ability of the mesenchymal stem cells and has stemness. The method of the present invention can verify the dryness of the mesenchymal stem cell gel derived from umbilical cord, fat, bone marrow and umbilical blood. |
