abstract | The invention discloses a method for constructing a high-efficiency expression vector by combining a high-CG segment and a low-CG-content promoter, wherein the method selects the promoter which contains an enhancer and has CG content less than 50% at the upstream of a transcription initiation site as a target promoter; replacing the CMV promoter in the exogenous gene expression frame on the commercial vector pEGFP-C1 by using the target promoter to construct an intermediate vector; inserting a CG fragment U1 with the CG content being more than 70% of 1-1.5kb into an Ase I enzyme cutting site before a foreign gene expression frame of the intermediate vector to construct an expression vector; the expression vector can ensure that the expression quantity of the EGFP gene in the expression frame of the target promoter can be continuously, stably and efficiently expressed by verification. The method can be applied to the industrial production field of antibody and protein medicines or gene therapy to improve the production efficiency and reduce the production cost, or can be applied to the field after further improvement. |
