abstract | The invention provides a culture method for forming early micro-hair follicles in vitro and its application. The culturing method includes: mixing dermal progenitor cells and epidermal stem cells, suspending culture in a medium to form dermal-epidermal aggregates, and collecting the cultured hair follicle-like structures to be cultured hair follicle-like micro-organs. This method enables the formation of a large number of hair follicle micro-organ structures (aggregates) within 24 hours only by a simple suspension method using scalp dermal progenitor cells and foreskin epidermal stem cells. At the same time, the in vivo transplantation experiments further confirmed that the in vitro cultured hair follicle micro-organs can form mature hair follicles in vivo. The present invention lays a good foundation for studying hair follicle epithelium-interstitial interaction, evaluating induced hair stem cells, screening those supporting hair follicle regeneration, and applying to clinical treatment of hair loss in the future. |
