titleOfInvention Recombinant penicillium chrysogenum gene engineering bacterium and construction method and application thereof
abstract The patent discloses a recombinant penicillium chrysogenum genetic engineering bacterium and a construction method and application thereof, wherein a sequence of a Pc-somA gene in the penicillium chrysogenum genetic engineering bacterium is replaced by a Bleo resistance gene; the penicillium chrysogenum is ATCC 48271; the construction method comprises the following steps: (1) extracting genome DNA of penicillium chrysogenum; (2) constructing a gene knockout fragment; (3) preparing protoplasts of penicillium chrysogenum; (4) and (3) introducing the gene knockout fragment into the prepared protoplast for homologous recombination to obtain the recombinant penicillium chrysogenum genetic engineering bacterium. Has the advantages that: the genetic engineering bacteria provided by the invention increase the biofilm amount OD570 of penicillium chrysogenum in the immobilized fermentation penicillin production process by nearly 2 times, so that hypha can be better adsorbed and combined with a plastic medium, the titer of penicillin is improved to 1963.25U/mL, the whole fermentation period is shortened, 24h is shortened, and the production cost is reduced.
