The present invention increases the pH of the medium by changing the carbon source of the medium and adding salt substances to neutralize the lactic acid produced during the culture, thereby improving the cold storage rate of the lactic acid bacteria. After comparative research, it was found that the modified MRS medium with sucrose instead of glucose had no adverse effect on the culture effect of lactic acid bacteria that could utilize sucrose, but could significantly reduce the mortality rate of refrigerated storage. 1% calcium carbonate was added to the culture medium during cultivation, and the preservation effect was better. The carbon source utilized in the present invention is sucrose, which is low in cost and easy to utilize. The neutralizing salt of the acid is calcium carbonate, which is small in amount and low in cost.
