abstract | The invention discloses a method for identifying a plurality of pairs of biomolecular interaction regulating factors in a biomolecular chain. On the basis of establishing a phase change system by using multivalent interaction macromolecules, the invention creates recombinant biomolecule chains, molecules among the recombinant biomolecule chains are gathered in phase change liquid drops through the interaction of ring-ring buckling, and whether the interaction exists between biomolecule pairs formed between upstream and downstream molecules of the recombinant biomolecule chains can be determined by marking a reporter group on a biomolecule at the chain end and detecting whether the gathering of a signal of the reporter group exists in the phase change liquid drops. On the basis, the biomolecule interaction inhibitor is added into the system, and the synchronous high-throughput screening of the multi-target biomolecule interaction inhibitor is realized by detecting the change of the signal intensity of the reporter group in the phase-change liquid drop. The method has the advantages of simple operation, high sensitivity, low cost and wide applicability, and is particularly suitable for screening signal channel regulators. |
