abstract |
The invention discloses an expression plasmid, a cell line for packaging second-generation adenovirus and its application. The cell line of the present invention was deposited in the China Center for Type Culture Collection on May 8, 2019, and the deposit number is CCTCC NO: C2019111, and the classification name is human embryonic kidney transformed cell AY293-6015. The cell line of the present invention comprises the E4-ORF6/7 gene of adenovirus, and can be used to package the second-generation adenovirus with the deletion of the E4 gene to form complete and infectious second-generation adenovirus particles. The eukaryotic plasmid of the present invention replaces the CMV promoter commonly used in viral vectors with the RSV promoter, thereby reducing the occurrence probability of homologous recombination in virus cell packaging and reducing the formation of RCA. |
