abstract | The invention discloses a novel preparation method of 5-acetylcytosine, which comprises the steps of taking cytosine as a raw material, adding the cytosine into a solvent, stirring, adding 3-5 equivalents of an acid reagent, adding 1-5 equivalents of an acetylation reagent, and heating to react to obtain the 5-acetylcytosine, wherein the acetylation reagent is acetyl chloride, acetyl bromide or acetic anhydride; the acidic reagent is Lewis acid or other acidic reagents such as phosphoric acid and sulfuric acid; the solvent is chloroform, dichloromethane, acetonitrile, N-dimethylformamide, toluene, nitrobenzene or dioxane. The method has the advantages of short reaction route, cheap and easily obtained raw material cytosine, mild reaction conditions, high conversion rate, suitability for industrial production and the like. |
