Disclosed herein are compositions, methods, and kits comprising engineered reverse transcriptases that exhibit several desirable properties, such as thermostability, processive reverse transcription, non-templated base addition, and template switching capabilities. The engineered reverse transcriptases described herein show unexpectedly higher resistance to inhibition by cell lysates, greater ability to capture full-length mRNA transcripts compared to other engineered reverse transcriptases , and show improved results in the case of small reaction volumes. |
