abstract The invention discloses a tumor microenvironment response type nuclear targeting platinum nanoparticle, which comprises a platinum nanoparticle, and a nuclear targeting peptide and an amphiphilic polymer which are modified on the platinum nanoparticle. The invention also discloses a preparation method of the tumor microenvironment responsive nuclear targeting platinum nanoparticles, which comprises the following steps: dissolving a platinum salt precursor and an end-capping reagent in water to form a precursor solution, and adding a strong reducing agent to carry out a reduction reaction to obtain ultra-small platinum nanoparticles; dissolving amphiphilic polymer in a good solvent, adding a modifier, and stirring for reaction to obtain amphiphilic polymer; dissolving the subminiature platinum nanoparticles and the nuclear targeting polypeptide in water, and stirring for reaction to prepare nuclear targeting platinum nanoparticles; dissolving the nuclear targeting platinum nanoparticles and the amphiphilic polymer in water, and stirring for reaction to obtain the tumor microenvironment response type nuclear targeting platinum nanoparticles. The tumor microenvironment response type nuclear targeting platinum nanoparticles can specifically target tumor cells, enter cell nuclei to damage DNA and kill the tumor cells.
