abstract | The invention discloses a p53 protein and mitochondria double-labeling immunofluorescence detection method and a kit thereof, wherein the detection method comprises the following steps: preparing a cell climbing sheet and cleaning; fixing with cell fixing liquid, and cleaning; sealing with cell sealing liquid; adding primary antibody for incubation and rinsing; the primary antibody is an antibody which is combined with p53 protein and an antibody which is combined with mitochondria; adding a fluorescence-labeled secondary antibody for dyeing, incubating and cleaning; re-dyeing and cleaning cell nucleus fluorescent dye; sealing, and observing a dyeing result by using a fluorescence microscope for microscopic examination or confocal microscope for microscopic examination; the fluorescence image superposition analysis only breaks the cell membrane without breaking the nuclear membrane, can avoid the signal interference in the nucleus, realizes the co-localization of the p53 protein and the mitochondria, and provides basis and reference for the study of the localization of the mitochondria of the protein and the localization of the protein and other non-nuclear organelles. |
