abstract | The invention provides a method and a target for screening JEV resistance genes by using CRISPR/Cas9 knockout library technology. Specifically, the invention provides a method for screening JEV resistance-related genes based on a whole genome CRISPR/Cas9 knockout library strategy and a candidate target gene. According to the method, a sgRNA plasmid library containing a pig complete genome is used for packaging lentivirus, a pig somatic cell stably expressed by Cas9 is infected, and a complete genome gene knockout cell library is obtained through enrichment by flow cytometry. And infecting a knockout cell library by using JEV, collecting the survival cells, extracting genome DNA, carrying out PCR amplification, establishing a library and high-throughput sequencing, and finally analyzing by using bioinformatics to obtain tens of JEV resistance candidate functional genes. The invention establishes a method for screening the JEV disease-resistant gene of a pig based on a whole-gene CRISPR/Cas9 knockout library strategy, and the identified candidate gene target can be used for targeted drug development and disease-resistant breeding research for inhibiting JEV replication. |
