abstract The present invention provides a cassava CAMTA gene and the construction and disease-resistant application of an inhibitory expression vector thereof. A gene is isolated from cassava, and the nucleotide sequence of its CDS is shown in SEQ ID No. Construction of Cassava CAMTA Gene Suppression Expression Vector: RNA from cassava leaves of Huanan No. 124 was extracted and reverse transcribed into cDNA, and the 1-600bp fragment at the 5' end of CAMTA gene was used as the specific nucleic acid silencing fragment of the virus-induced gene silencing system to obtain cDNA. A 1-600 bp fragment at the 5' end of the CAMTA gene is obtained by amplifying the template, and then the gel is recovered and then connected to the TRV virus-induced gene silencing vector pTRV2 to obtain the pTRV-MeCAMTA gene silencing expression vector. By constructing the gene silencing expression vector and injecting cassava leaves, an effective transient MeCAMTA gene silencing cassava was obtained, which provided an effective method for studying gene function.
